Description
We are pleased to introduce two new additions to the Strict-R™ inducible CRISPR platform designed to deliver stringent OFF-state control and robust ON-state activity for gene knockout (CRISPRko) and gene repression (CRISPRi). These systems use a dual-layer regulatory design combining Tet-On 3G transcriptional control with an FKBP12-derived destabilizing domain, ensuring Cas9 or dCas9 fusion proteins remain tightly suppressed until both doxycycline and Shield1 are added. These two additions complete support of all Dharmacon reagents CRISPR systems by complementing the current Strict-R CRISPRa offering for gene activation.
